Abstract Bauhinia forficata Link aqueous extract is usually recommended as a phytomedicine to reduce blood glucose levels and its biological activity has been linked to the presence of phenolic compounds from B. forficata preparations. Several drying processes are used in the production of dry herbal extracts, which may influence the chemical composition and efficacy of final herbal medicines. Due to significant chemical changes, defining appropriate drying processes is essential for phytopharmaceutical drug development. In view of this, we analyzed dried B. forficata leaf infusion (BFLI) extracts by HPLC-UV-MSn, followed by molecular networking analysis to evaluate the chemical profiles from dried extracts yielded by freeze-and spray-drying processes. The main metabolites detected included 11 ferulic/isoferulic acid derivatives and 13 glycosylated flavonoids. The qualitative chemical profiles were alike for both drying processes, whereas the relative abundance of some flavonoids was higher using spray-drying. Taken together, our results showed that freeze-and spray-drying preserved the phenolic profile of BFLI and suggested that spray-drying may be the most suitable to obtain its dried products. Along with studying the chemical profiles of dried herbal extracts, evaluating the influence of drying processes on the quality and chemical profiles of final products is pivotal and may benefit future research.

The genus Lychnophora (Asteraceae) is a medicinal plant traditionally used as anti-inflammatory or analgesic agent. Its biological properties have been widely investigated and correlated with its chemical composition, which could be different between the species and even accordingly to the place and season of collection. In this work, it was investigated the spatial and temporal variations in the secondary metabolites from five populations of Lychnophora ericoides. Among them, a more diversified secondary metabolism was observed for the plants collected at Ibiraci-MG (Brazil), a city placed between two biomes, Cerrado and Atlantic Forest. The highest concentrations of chlorogenic acids were found in the Cerrado dry season (winter), while the flavonoids were higher during the rainy season (summer). Significant reduction was observed in the contents of all the secondary metabolites in old leaves, and also in flowery branches. These variations detected in L. ericoides leaves could highly influence the intensity of the biological activities of this plant.

The species Eremanthus mattogrossensis, known as "veludo do cerrado" (cerrado velvet), is native to the Brazilian Cerrado. Because the amount of metabolites present in plants may be influenced by biological and environmental factors, here we conducted an HPLC-DAD-MS/MS investigation of the metabolite concentrations found in the MeOH/H2O extract of the leaves of this species. The main compounds were identified and quantified, and the metabolites were grouped by chemical class (caffeoylquinic acids, flavonoids, and sesquiterpene lactone). Statistical analysis indicated a straight correlation between the quantity of metabolites and seasonality, suggesting that environmental properties elicit important metabolic responses.

Este artigo descreve a caracterização por CLAE-DAD-EM/EM e CLAE-DEC dos perfis de metabólitos secundários de extratos das folhas de Lychnophora ericoides Mart. de diferentes procedências e suas propriedades citotóxicas e antioxidantes. Extratos foliares de populações representando sete diferentes localidades foram avaliados quanto à atividade antioxidante pelo sistema DPPH e à atividade citotóxica em três linhagens tumorais. Os extratos foram eletroquimicamente caracterizados por CLAE-DEC e seus metabólitos secundários majoritários foram identificados por CLAE-DAD-EM e CLAE-DAD-EM/EM. Um metabolismo secundário defensivo amplificado, juntamente com as mais altas bioatividades antioxidantes e citotóxicas, foi encontrado para as plantas coletadas na interface entre dois tipos de vegetação. Estes resultados fornecem apoio adicional para a hipótese de que plantas ocorrendo na interface entre dois ecossistemas podem ser estimuladas a amplificar sua produção e armazenamento de metabólitos secundários defensivos, devido ao maior número de influências ambientais.

This paper reports HPLC-DAD-MS/MS and HPLC-ECD characterisation of secondary metabolite profiles of Lychnophora ericoides Mart. leaf extracts from different provenances and their cytotoxic and antioxidant properties. Leaf extracts from populations representing seven different locations were evaluated for antioxidant activity by the DPPH radical scavenging system and activity towards cellular growth in three tumor cell lines. The extracts were electrochemically analysed by HPLC-ECD and their main secondary metabolites were identified by HPLC-DAD-MS and HPLC-DAD-MS/MS. An amplified defensive secondary chemistry, together with maximal cytotoxic and antioxidant bioactivities, were found for plants collected at the interface between two types of forest. These findings furnish additional support for the hypothesis that plants occurring at the interface between two forests ecosystems might be stimulated to amplify their own production and storage of defensive secondary metabolites due to the greater number of environmental influences.

This paper describes the antioxidant effects of thirteen phenolic compounds isolated from plants of the genus Lychnophora. Two assays were performed to evaluate these effects: a cellular test that measured the luminol-enhanced chemiluminescence produced by neutrophils stimulated with opsonized zymosan and a cell-free test involving horseradish peroxidase-H2O2-luminol. In both assays, the antioxidant activity of the phenolic compounds was dependent on their concentration and chemical structure. Our results suggest that the ability of phenolic compounds from Lychnophora species to scavenge and inhibit the generation of ROS may be a mechanism underlying the anti-inflammatory activity of extracts from Lychnophora spp.

Since secondary metabolites represent a chemical interface between plants and surrounding environment, their syntheses are frequently affected by environmental conditions. Thus, variations in the total content and/or of the relative proportions of secondary metabolites in plants can take place. We review the main environmental factors that can streamline or alter the production or concentration of secondary metabolites in plants. How seasonality, circadian rhythm, developmental stage and age, temperature, water availability, UV radiation, soil nutrients, altitude, atmospheric composition and tissue damage influence secondary metabolism are discussed.

Eremanthus seidelii MacLeish & Schumacher tem ocorrência restrita ao cerrado, em torno do reservatório de Furnas (MG), em habitats que vêm sendo seriamente deteriorados pela ação humana. A presente investigação fitoquímica mostrou, como metabólitos secundários majoritários das folhas de E. seidelii, as lactonas sesquiterpênicas (LS) 4beta,5-diidro-2',3'-diidroxi-15-desoxi-goiazensolídeo (1) e 4beta,5-diidro-1',2'-epoxi-eremantolídeo-C (2); uma metodologia foi desenvolvida em CLAE para sua análise quantitativa. A análise por CLAE mostrou que não há variações sazonais significativas nas concentrações de ambas LS. Não foram encontradas diferenças qualitativas nos perfis de LS nos indivíduos amostrados. Entretanto, há diferenças no perfil quantitativo entre os indivíduos analisados, apontando para a existência de três quimiotipos quantitativos desta espécie, com diferenças possivelmente originadas na atividade enzimática das enzimas que ciclizam uma LS tipo goiazensolídeo (1) em uma LS tipo eremantolídeo (2).

Eremanthus seidelii MacLeish & Schumacher has a restricted occurrence to the Brazilian "cerrado" surrounding the Furnas (MG) reservoir, in environments that have been seriously damaged by human activity. The present phytochemical investigation reveals that the sesquiterpene lactones (SL) 4beta,5-dihydro-2',3'-dihydroxy-15-desoxy-goyazensolide (1) and 4beta,5-dihydro-1',2'-epoxy-eremantholide-C (2) are the major secondary metabolites in E. seidelii leaves, and an HPLC method was developed for their quantitative analysis. HPLC analysis showed no significant seasonal variation in the concentrations of both SL. No qualitative differences were found in the SL patterns of all individuals sampled. However, there is a different SL quantitative pattern among the plants analyzed, pointing to the existence of three quantitative chemotypes of this species, with differences possibly originating from the activity of the enzymes that cyclize the goyazensolide type SL (1) to a eremantholide type SL (2).
